Opinion or fact?
Scorpion and Subzero have been around the longest, appearing in every MK game.
I, personally, think Smoke is the best, though.
As for DC, definitely Batman and Superman.

The best matters on the person playing mine are Reiko and Sareena but it can very from person to person what type of best do you mean best fighter, best story for a fighter or what?

I really don't understand how you can just ask who the "best" character is. There's a lot of things to consider. I mean, there's appearance, powers, knowledge, favoritisms, etc. I think that people who reply to this thread are just going to choose their favourite people from each universe. So this is what I'm gonna do.
Mortal Kombat: Shao Kahn
I still don't completely understand why I like Shao Kahn so much. I think that I just like the fear he puts into everyone. No-one f*cks with him. He is one of the most powerful and feared characters throughout the realms. He also has a cool look {in the later games, especially} and he is THE boss. He is the boss of bosses. In the words of John Vogel, he is the "top of the ladder".
DC Universe: Batman
I don't know why I quite like Batman. I think I'm guilty of only liking him because he is so iconic in the DC comic universe. I also admire the fact that even though he's a billionaire, he still devoted to destroying crime in Gotham City. I also admire how he's the only "superhero" who has no powers whatsoever, but he's still one of the most iconic characters of the DC universe.

That was uncalled for dude. I think that just because someone does not write/post using correct spelling and/or grammar, that doesn't mean he is in elementary school, or even dumb. I know a guy who is in high school, and gets straight A's, but still posts using bad grammar because he's very lazy. Still though, I don't feel that being rude to Darkeblademaster is the way to go.

i agree. sometimes i dont spell words right but its because im too lazy to spell things right lol. or i just write things without looking at it and just post it without correcting it.
